Can pyrroloquinoline quinone (PQQ) protect chilled ram sperm well enough to improve real artificial-insemination outcomes?
A 2027 study in Theriogenology paired in-vitro sperm assays with a large field trial: 800 ewes timed cervical AI using semen stored at 4°C, with PQQ added to the extender at 10 / 100 / 1,000 / 10,000 nM versus control.
Why this matters Ram sperm membranes are rich in polyunsaturated fatty acids. Cold liquid storage triggers oxidative stress—ROS surge, motility drop, membrane and mitochondrial damage—and conception suffers. Antioxidants in extenders are common; PQQ is different from classic VC/VE: redox-cycling (one molecule can quench many radicals), mitochondrial support, and Nrf2 pathway activation. Prior work was mostly in vitro. This study connects sperm metrics to pregnancy and lambing.
What the data show
→ Optimal dose: 100 nM. After 24–48 h storage, 100 nM beat control on total motility, progressive motility, viability, mitochondrial membrane potential, and membrane integrity—while MDA (lipid peroxidation) fell. → Hormesis alert: 10,000 nM was pro-oxidant—sperm metrics worse than control. 10 nM and 1,000 nM helped somewhat, but less than 100 nM. → Field fertility: Cold-stored control conception ~29%; 100 nM PQQ ~55% conception and ~57% lambing—nearly doubled, yet still below fresh-semen benchmarks. Twin rates did not differ.
Mechanism & production takeaways Direct ROS scavenging + mitochondrial complex stabilization + Nrf2-driven endogenous enzymes. Antioxidant additives have a narrow dose window—overdose can reverse the benefit. This protocol is for 4°C liquid ram semen, not freeze–thaw; cross-species and human translation remain open questions.
Caveat In-vitro sperm improvement can translate to field fertility, but it is not equivalent to fresh semen—and animal data do not prove a human male-fertility claim.
