Recently, a research team from Zhejiang University published a study in the Journal of Advanced Research, which systematically revealed for the first time the key rate limiting steps of plant sterol absorption and its synergistic mechanism with dietary monounsaturated fatty acids (MUFA).<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Food sources of plant sterols<\/h2>\n\n\n\n<p>Phytosterols are widely present in daily plant-based foods. **Plant oil is the most abundant source, especially corn oil, soybean oil, sesame oil, and olive oil. **Nuts and seeds such as sesame, sunflower seeds, almonds, peanuts, etc. are also abundant. In addition, legumes, whole grains, and fruits and vegetables also contain a certain amount of plant sterols. It is worth mentioning that plant sterols often naturally coexist with MUFA in the same type of food, such as olive oil, avocado, almonds, etc., providing evidence for the synergistic mechanism discovered in research.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Key finding: Esterification is the true limiting step<\/h2>\n\n\n\n<p>To reveal the true rate limiting process of plant sterol absorption, the research team used a combination of in vitro simulated digestion and in vivo animal experiments to systematically evaluate the absorption process of plant sterols.<\/p>\n\n\n\n<p>Research has found that even if the ratio of plant sterols to cholesterol is increased to 10:1 (far beyond the actual intake ratio), the solubility of plant sterols in mixed micelles is significantly improved, but the esterification efficiency gradually increases from 60% to 90%. This result indicates that when plant sterols enter intestinal cells, their esterification efficiency is the key factor determining absorption.<\/p>\n\n\n\n<p>**ACAT2 (Acyl CoA: Cholesterol Acyltransferase 2) * * is the core enzyme that catalyzes esterification reactions. After using ACAT2 inhibitors, the levels of cholesterol esters and phytosterol esters in the blood significantly decreased, confirming that ACAT2 is essential for the esterification of both types of sterols. Therefore, ACAT2 mediated esterification is the fundamental rate limiting step in the absorption of plant sterols.<\/p>\n\n\n\n<p>It is worth noting that ACAT2 catalyzed esterification requires acetyl CoA and oleoyl CoA as substrates, and the synthesis of these coenzymes is closely related to cellular mitochondrial energy metabolism. Maintaining healthy mitochondrial function, such as promoting mitochondrial biogenesis through pyrroloquinoline quinone (PQQ), may provide sufficient energy and substrate support for the esterification process in intestinal cells.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Dual regulatory mechanism of dietary MUFA<\/h2>\n\n\n\n<p>After clarifying esterification as the key rate limiting step, the research team further explored how MUFA affects this process. They found that MUFA synergistically enhances the esterification of plant sterols through a dual mechanism:<\/p>\n\n\n\n<p>**1. Variable conformation regulation: Through molecular docking and dynamic simulation, the molecular mechanism of the binding between acyl CoA (MUFA metabolite) and ACAT2 has been revealed. Oil acyl CoA forms a stable ternary complex with plant sterols in the ACAT2 catalytic pocket, reducing the binding free energy of plant sterols by 30% and significantly enhancing the enzyme&#8217;s affinity for plant sterols. In vitro enzyme activity assays confirmed that ACAT2 preferentially esterifies plant sterols in the presence of MUFA, rather than cholesterol.<\/p>\n\n\n\n<p>**2. Transcriptional regulation * *: MUFA activates the nuclear receptor FXR, initiating a transcriptional repression pathway. FXR activation inhibits the expression of transcription factor CDX2, which is a positive regulator of ACAT2 gene, leading to downregulation of ACAT2 expression. This seemingly contradictory phenomenon is essentially a reprogramming of substrate preferences &#8211; a reduction in enzyme quantity leads to a decrease in cholesterol esterification, but MUFA enhances catalytic efficiency through conformational changes, making the remaining enzyme molecules more inclined to esterify plant sterols.<\/p>\n\n\n\n<p>From the perspective of epigenetic regulation, the normal operation of this transcription pathway involves multiple methylation modification processes. **S-adenosyl-L-methionine (SAM-e) plays a fundamental role as an important methyl donor in maintaining the homeostasis of gene expression regulation in the body.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Metabolic reprogramming and its clinical significance<\/h2>\n\n\n\n<p>In addition to directly regulating ACAT2, MUFA also reshapes the metabolic environment of the intestine. A high MUFA diet significantly increased the levels of acetyl CoA and oleoyl CoA in the intestine, providing sufficient substrates for esterification reactions. At the level of fatty acid metabolism, research has confirmed the optimization effect of oleic acid on the catalytic conformation of ACAT2. It is worth further exploring whether similar structural functional relationships exist in other unsaturated fatty acids, such as DHA (docosahexaenoic acid). As an important source of omega-3 polyunsaturated fatty acids, DHA algal oil also has potential in regulating intestinal lipid metabolism.<\/p>\n\n\n\n<p>In addition, the study observed that a high MUFA combined with plant sterols diet has a protective effect on the intestinal mucosal structure of mice. In terms of maintaining intestinal health, * * Vitamin K2 * * also has a potential supportive effect on maintaining the homeostasis of intestinal epithelial cells by regulating calcium metabolism and osteocalcin activity.<\/p>\n\n\n\n<p>This study systematically elucidated the rate limiting steps of plant sterol absorption and revealed the molecular basis of MUFA enhancing plant sterol esterification through a dual mechanism. These findings have important clinical translational value: pairing foods rich in plant sterols (such as vegetable oils and nuts) with foods rich in MUFA (such as olive oil and avocados) can synergistically enhance cholesterol lowering effects. Fortunately, many natural foods themselves contain both of these ingredients, making them ideal daily choices.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Conclusion<\/h2>\n\n\n\n<p>This study by Zhejiang University not only deepens the understanding of gut sterol metabolism, but also provides scientific basis for optimizing the cholesterol lowering application of plant sterols.
